My thesis research will be focused on potential therapeutics to decrease motivation to take cocaine in a self-administration rodent model. My focus will be on finding a combination therapy to combat the dysregulations of the dopamine and dynorphin system seen after chronic cocaine exposure. During abstinence, for instance, a negative affect persists in cocaine-dependent individuals; this subsequent reward deficiency, a heightened stress response, and an inability to feel pleasure often leads to relapse. The cycle from abstinence to withdrawal to subsequent escalation of drug use is far too common, and even though ample therapeutics targeted at the dopamine and dynorphin systems have been created, none have yet been proven effective and approved by the FDA to treat cocaine addiction.
